Inositol is a cyclic polyhydroxy alcohol belonging to the group sugar alcohol, occurring in nine different spatial conformations, of which the most important is the meso isomer. Natural sources of this substance are oilseeds and leguminous plants, as well as oranges and peanuts.Although being traditionally known as vitamin B8, inositol is technically not a vitamin because of the fact that it is also synthesized by the human body and does not need to be provided with food. Inositol is used by the body to synthesize inositol triphosphate (IP3) and diacylglycerol (DAG), which act as second messengers of information within a cell.2

It affects the overall level of serotonin and GABA receptor binding resulting in an improved mood. It also gives a sense of a free flow of ideas and a sense of peace, allowing for better coping strategies with everyday stress.3 Studies using high doses of inositol have shown its effectiveness in cases of depression and anxiety disorders (social phobias, obsessive-compulsive disorders). Patients who were administered 18g/day of inositol for a duration of 6 weeks demonstrated a significant mood improvement. This effect was comparable with the effects of therapies utilizing SSRI (selective serotonin reuptake inhibitors) drugs, which are currently used in the treatment of depression.4,5 Furthermore, it was demonstrated that inositol is effective in the treatment of bipolar disorder.6


On average, 1 g of inositol is provided to the body from food sources. In cases of depresion or anxiety disorders, doses of 12-18 g/day divided into 2-3 smaller doses are used.4,5


Inositol has a strong synergic effect with substances that regulate acetylcholine levels such as choline, sulbutiamine, or DMAE. The combination with sulbutiamine or aniracetam produces an anti-depressant effect. Additionally, inositol can be combined with caffeine or L-carnitine.3

Side effects

Inositol is well tolerated even at doses of 18 g/day. May cause nausea, diarrhea, dizziness and headache.7
